چکیده

Duplicated sequences are substrates for the emergence of new genes and are an important source of genetic instability associated with rare and common disease. Analyses of primate genomes have shown an increase in the proportion of interspersed segmental duplications within the genomes of humans and great apes. This is in contrast to other mammalian genomes that appear to have their recently duplicated sequences organized in a tandem configuration. We discuss the mechanistic origin and the impact of this difference with respect to the evolution, genetic diversity and primate phenotype. While many genomes will be sequenced in the future, resolution of this aspect of genomic architecture still requires high quality sequence and detailed analyses.

Serial segmental duplications during primate evolution result in complex human genome architecture.

The human genome is particularly rich in low-copy repeats (LCRs) or segmental duplications (5%-10%), and this characteristic likely distinguishes us from lower mammals such as rodents. How and why the complex human genome architecture consisting of multiple LCRs has evolved remains an open question. Using molecular and computational analyses of human and primate genomic regions, we analyzed the...

An Alu transposition model for the origin and expansion of human segmental duplications.

Relative to genomes of other sequenced organisms, the human genome appears particularly enriched for large, highly homologous segmental duplications (> or =90% sequence identity and > or =10 kbp in length). The molecular basis for this enrichment is unknown. We sought to gain insight into the mechanism of origin, by systematically examining sequence features at the junctions of duplications. We...

Analysis of segmental duplications via duplication distance

MOTIVATION Segmental duplications are common in mammalian genomes, but their evolutionary origins remain mysterious. A major difficulty in analyzing segmental duplications is that many duplications are complex mosaics of fragments of numerous other segmental duplications. RESULTS We introduce a novel measure called duplication distance that describes the minimum number of duplications necessa...

The origins of genomic duplications in Arabidopsis.

Large segmental duplications cover much of the Arabidopsis thaliana genome. Little is known about their origins. We show that they are primarily due to at least four different large-scale duplication events that occurred 100 to 200 million years ago, a formative period in the diversification of the angiosperms. A better understanding of the complex structural history of angiosperm genomes is ne...

Segmental Duplications as a Complement Strategy to Short Tandem Repeats in the Prenatal Diagnosis of Down Syndrome

Background: Quantitative fluorescence-polymerase chain reaction (QF-PCR) is an inexpensive and accurate method for the prenatal diagnosis of aneuploidies that applies short tandem repeats (STRs) as a chromosome-specific marker. Despite its apparent advantages, QF-PCR is not applicable in all cases due to the presence of uninformative STRs. This study was carried out to investigate the efficienc...
